After setting up your video source under "Video source," you need to go 
into "Input connections" and assign your new video source to an input on 
each tuner card.  It's not enough to just set up the capture cards 
themselves.

In my case, I have my video source (analog cable) assigned to Tuner 0 on 
each of my PVR-250s (/dev/video0 and /dev/video1).
