Spotify is not a web based service you need to use a client (Windows or Mac
but I'm running it quite happily under Wine and they even provided
instructions for doing so).

They do provide an api http://developer.spotify.com/en/libspotify/overview/

"The libspotify C API package allows third party developers to write
applications that utilize the Spotify music streaming service. Hopefully,
this will enable and inspire you to build some really cool stuff. We’re
looking forward to seeing what you can come up with."

I've not studied the terms
http://developer.spotify.com/en/libspotify/terms-of-use/ but from their
description it seems likely that a mythplugin would be allowable.

There has been lot's of speculation about when Spotify is launching in the
US and my reading of it is that it is "when" rather than "if"

I think Spotify is a great service and I'm a happy paying customer.
