No, you can do that at any time but it goes away at each reboot.

Here is what I have on 1 server at work:

echo "1024" >> /sys/block/md0/md/stripe_cache_size
echo "1024" >> /sys/block/md1/md/stripe_cache_size

md0 is a 10 drive (250GB sata I) raid 6
md1 is a 5 drive (250GB sata I)  raid 5

BTW, 2048 is probably too high in most cases.
