Umm... not really. A serial port is a 9 pin connector and does not
look like a phone jack. What you are probably thinking of is the IR
that comes with some capture cards. But yes, you can control MythTV
with with either a homemade serial cable or one that comes with some
capture cards.
As for the PVR-250 vs. PVR-350 debate, my vote is for the 250 and a
good video card.
