Use the physical dimensions of the screen to determine the aspect  
ratio.  Nothing to do with pixels.  Or, alternatively, determine DPI of  
X and Y separately, then figure out how many pixels you need given that  
DPI.  However you want to think of it.

Some examples of non-square pixels to keep in mind as you do this...  
some HDTV projectors are 1024x1024 pixels, yet are 16:9 in physical  
dimensions.  And some HDTV CRTs are (can be run at) 1920x540, yet are  
16:9 in physical dimensions.  Also, your 1280x1024 example... it's 4:3  
in physical dimensions.
